Describe DNA Replication. 

DNA replication begins at the origin of replication. Helicase unwinds the double helix structure by breaking the hydrogen bonds between the complementary base pairs in the parental strand. [1]


Each parental strand acts as a template for the synthesis of a new daughter strand. Primase attaches itself to the unwound strand and catalyses the synthesis of RNA primers which help to provide free 3’OH ends for DNA polymerase III. [1]


DNA polymerase III then catalyses phosphodiester bonds between the free incoming deoxyribonucleotides and free 3 ‘OH ends, lengthening the daughter strand in the 5’ to 3’ direction. Daughter strand links itself to the parental strand by complementary base pairing, with A to T and G to C. [1]


Leading strand is synthesised continuously while lagging strand is synthesised discontinuously, forming Okazaki fragments. [1]


RNA primers are removed and replaced by deoxyribonucleotides by DNA Polymerase I. Nicks between Okazaki fragments are filled by DNA ligase through forming phosphodiester bonds. [1]
