Describe the process of creating a MAB

1.Scientists identify and isolate the antigen from target cell (ex: cancer cell)

2.An animal (usually mouse) is vaccinated with the antigen, which stimulates the selection and proliferation of B cells specific for the antigen

3.B cells are extracted from the spleen of the animal

4.B cells are fused with myeloma cells (rapidly dividing cancerous cells) to produce hybridomas. This is to promote B cell growth quickly and in large quantities

5.Hybridomas are screened so that only cells with the appropriate antibody are selected

6.Hybridomas are cloned and produce mass amounts of antibodies

7.Antibodies are collected, purified and used to treat the patient

Explain the process of attenuation

When there is low availabilty of trp (only on tRNA), as RNA polymerase transcribes the trp operon regulatory genes and the ribosome translates the mRNA, stopping at the STOP codon, then a section of the mRNA forms an attenuation loop, causing RNA polymerase to be pulled off the trp operon, ending translation, therefore no trp is made.
