Macbeth seems sincere in his intentions in the first scene; when he says farewell to Banquo “I wish your horses swift and sure of foot, and so I do commend you to their backs”(III.i.38-39). Banquo doesn’t have a clue that he has a target on his back. Macbeth paints Banquo as an enemy to the Murderers-Banquo ruined their and their families lives ; in truth, Banquo has done nothing to them, Macbeth was probably the one who caused hardship, “That it was he, in the times past, which held you so under fortune, which you thought had been our innocent self”(III.i.77-79) .

Lady Macbeth doesn't encourage Banquo's murder. Normally Lady Macbeth would have encouraged Macbeth to kill anyone standing in Macbeth's way.
