If I washed the blood off multiple times but there is still a stain, would my prayer count then?
No, your prayer would not count because blood is Najasah. You clean it by removing the excess blood off the clothes then running water through it and washing it.
Your prayer would count then because you washed it so everything is gone except for the stain.
Are fish Najs? If yes, how do we purify them, if not why?
They are not Najs, because anything that comes out of the water is halal for us = not najs
