what does hcc stand for?
hepatocellular carcinoma
what does dicer do?
cleaves pre-miRNA near the terminal loop
what type of ago do we use?
ago2 (there is ago1-4 in humans)
what micropipette would i use for 20 ul? 2 ul? 150 ul?
20 ul pipette, 2 ul pipette, 200 ul pipette
how can i find where a specific chemical is?
search on chem inventory!
how is it diagnosed?
ultrasound, ct scan, mri, biopsy (expensive and invasive procedures
what does trbp do?
transports the mature mi-RNA to the risc complex
what is the full name of trbp?
trans-activation response rna binding protein
how much amp would i need for a 5ml dicer culture?
10 ul
if you need help while in lab, what should you do? ex. confused on a procedure
call one of us!! or ask someone else in lab if ur brave
how common is hcc?
75-85% of liver cancer
what does ago do?
main component of risc complex, helps mi-RNA bind to target mRNA
what is the full name of the risc complex?
rna-induced silencing complex
(bonus points if they know which type of silencing it is)
how do you prepare to use the laminar flow hood?
how do you dispose of bacterial cultures?
bleach them, then pour them down the drain
what other diseases are hcc associated with?
hepatisis b and c
why do we use miRNA-122?
low levels of miRNA-122 have been associated w hcc since miRNA-122 is a tumor suppressor and helps regulate apoptosis (cell death so cells don't grow out of control and become cancerous)
what is the size of dicer?
~219 kDA
what temp are antibiotics, proteins, and bacterial cultures stored at?
where are 3 places a chemical may be?
bio, chem, chemical reagant room
why is hcc hard to diagnose?
liver is a large organ that compensates for damage until too late, early stage of hcc does not have any unique symptoms
how does miRNA help gene regulation and why is gene regulation important?
when miRNA binds to its target mRNA, it will either degrade the mRNA or inhibit translation (mRNA cannot become protein). gene regulation allows life to respond to its environment by expressing different genes. it also allows cells to specialize to the body's needs (ex. we don't want our eye cells to product stomach acid), so not all our cells are exactly the same even though they have the same DNA
what antibiotic does each protein use?
ago and dicer - ampicillin
trbp = kanamycin
what are the 3 medias we use for bacterial cultures (full names!)
luria broth (lb), terrific broth (tb), autoinduction
how long should tb cultures incubate for?
19-21 hours
